Abstract: The nicotinic acetylcholine receptor mediates rapid neurotransmission at the neuromuscular junction and other sites within the central and peripheral nervous system. Reliable, fast channel response to the binding of acetylcholine is critical for proper functioning. Kinetic measurements of binding to the acetylcholine sites show two-fold enhancements in association rate in agreement with the potential measurements. At the ion channel, however, computed potentials over-estimated the measured potentials substantially. This suggests a need for improved experimentation or computation to understand electrostatic interactions within the confines of ion channels. Further kinetic measurements reveal that binding regulates opening and subsequent desensitization of the channel predominantly through one of the two binding sites, the alpha-gamma site.
