Abstract: Neurotransmitter transporters present at the plasma membrane contribute to the clearance and recycling of neurotransmitters and have a profound impact on the extent of receptor activation during neuronal signaling. In addition to their role in neurotransmitter clearance, these carriers are the primary targets for psychostimulant drugs of abuse, therapeutic antidepressants and other drugs used to treat brain disorders. Our research efforts have focused on the structure, regulation and cellular physiology of two families of sodium-dependent neurotransmitter transporters, the biogenic amine and the excitatory amino acid carriers. Although these transporters have their primary role in neurotransmitter clearance, members of both families also display a ligand-gated chloride channel activity that can regulate neuronal excitability and neurotransmitter release. This lecture will consider neurotransmitter transporter functions and their role as drug targets and will present results from structural, electrophysiological and cell biological studies aimed at defining the relationships between neurotransmitter transporter structure, substrate transport, and ion permeation.
